An innovative approach for low input forensic DNA sample analysis using the Magelia® platform



This article explores a novel method for analyzing forensic DNA samples with low input using the GlobalFiler™ IQC PCR amplification Kit on the Magelia® platform.

Inorevia in collaboration with the Criminalistic Biology Genetics Division of the Institut de Recherche Criminelle de la Gendarmerie Nationale (IRCGN), conducted an innovative approach aiming to improve the analysis of forensic DNA samples when dealing with trace or degraded samples.

Pairing the GlobalFiler™ application with the unique features of the Magelia® platform including: full automation, reaction encapsulation in capillaries and optimized reaction volumes were at the heart of this study. This strategy showed distinct advantages over standard treatment, notably increased signal for lower DNA inputs. Importantly, negative casework samples through routine treatment yielded "usable" DNA profiles after amplification using this strategy, making it a valuable tool for forensic laboratories.

This approach addresses common challenges in forensic DNA analysis, such as low template DNA, the need to secure rapid results, and the capacity to archive left over material, thereby showing promise for its integration in forensic workflows.
